Tupelo Blues wurde im April 1959 aufgenommen und war Hookers erste LP für den traditionellen Folk / Blues-Markt, wo die Blues-Legende nichts anderes als die Akustikgitarre spielte und mit seiner seelendurchtränkten Gesangsdarbietung glänzte. Das Album stellte eine entscheidende Veränderung gegenüber seinen früheren Aufnahmen dar, auf denen Hooker größtenteils auf einer E-Gitarre mit seinem typischen Hall zu hören war. Dies ist eine intime Session mit Standards wie »How Long Blues«, »Bottle Up and Go« und seiner ersten aufgenommenen Version von »Tupelo Blues«, einem seiner Klassiker.
