Peter Parker ist ein normaler Junge, der als Waise bei Tante May und Onkel Ben aufwächst und obendrein für die schöne Nachbarstochter Mary Jane schwärmt. Als er jedoch bei einer Schul-Exkursion von einer genmanipulierten Super-Spinne gebissen wird, entwickelt Peter plötzlich unglaubliche Fähigkeiten: Phantastische akrobatische Talente, einen übernatürlichen Instinkt für Gefahren und die Begabung, Netze zu spinnen. Begeistert macht er das Beste aus seinem neuen Potential: Geld. Doch dann wird Onkel Ben bei einem Überfall getötet und sein Vermächtnis "Aus großer Kraft folgt große Verantwortung" lastet schwer auf ihm. Als SPIDER-MAN widmet Peter Parker nun sein Leben dem Kampf gegen das Böse. Sein Erzfeind kommt aus dem direkten Umfeld: Norman Osborn, Vater von Schulfreund Harry, mutiert zum skrupellosen Grünen Kobold und terrorisiert New York.
